Sample Submission & Data Presentation

It is recommended to submit samples to the Lipidomics Shared Resource in 15mL conical tubes for all cell, tissue homogenate, or other solid matrices, and if possible provide samples in 8 mL glass tubes or the 15 mL conical tubes for media, plasma, blood, or other liquids.

  • Consecutively label tubes (1 - ∞) along with any additional info needed.
  • Total amount of submitted sample will be used for extraction/analysis unless otherwise noted.
  • For cell samples, provide a minimum of 1 million cells for each sample. Pellet samples and remove any supernatant before submission.
  • For tissue samples, homogenize and determine total protein concentrations and submit a minimum of 1 mg of total protein from tissue homogenate per sample.
  • For blood, plasma, or serum provide 50-100 µL of sample.
  • For cell culture media, provide 2 mL of sample.

Analytical data is reported in a concentration of [pmole/total sample]. The use of data normalization values (Phosphate (Pi), Protein, or Volume) are used for final concentration units of sample values. The data normalization process and determination are responsibilities of the client. The Lipidomics Shared Resource will determine Phosphate (Pi) levels for outside MUSC clients at a charged service fee. For MUSC and Hollings Cancer Center clients, the Lipidomics Shared Resource will save an aliquot of the sample extract for the client’s pick-up and use in determining Pi levels.

Analytical results are to be expressed as one of the following:

  • Lipid level (pmole)/inorganic phosphate level determined from the Bligh & Dyer lipid extract (nmole).
  • Lipid level (pmole)/Protein level (mg).
  • Lipid level (pmole)/Volume (µL) reflecting in molar concentration [M].
  • Lipid level (pmole)/Tissue weight (mg).
